Terpinolene in Cannabis

What is Terpinolene?

Terpinolene (C₁₀H₁₆) is a terpene found in cannabis, apples, cumin, lilac and tea tree. In cannabis, it contributes to the characteristic aroma and may modulate pharmacological effects through the proposed entourage effect.

Aroma: Fresh, piney, floral and herbaceous — a complex multi-note aroma.

Pharmacological Properties

Less common as dominant terpene than myrcene or limonene but predominant in certain cultivars (notably Jack Herer and related varieties). Preclinical: CNS depressant (sedative in mice), antioxidant, antibacterial. Antiproliferative in cancer cell lines in vitro.

Relevance in Cannabis

Terpinolene-dominant cultivars are paradoxically associated with uplifting, energetic effects in qualitative reports — despite preclinical sedative findings. This discrepancy highlights the complexity of terpene-cannabinoid interactions and the limits of translating preclinical terpene pharmacology to clinical effects.

GMP and Manufacturing Relevance

Include in comprehensive terpene profiling for full-spectrum preparations. Terpinolene is included in the cannabis terpene panel of most validated analytical methods for pharmaceutical quality testing.

Analytical Measurement

Terpene profiling is performed by headspace GC-MS or GC-FID. Validated methods should achieve quantitative separation of key terpenes. Where terpene content is a product specification parameter, the analytical method must be validated under ICH Q2(R1).
