What is Beta-Caryophyllene (BCP)?
Beta-Caryophyllene (BCP) (C₁₅H₂₄) is a terpene found in cannabis, black pepper, cloves and hops. In cannabis, it contributes to the characteristic aroma and may modulate pharmacological effects through the proposed entourage effect.
Aroma: Spicy, peppery, woody — the aroma of black pepper is primarily BCP.
Pharmacological Properties
Unique among terpenes as a functional CB2 receptor agonist — the only known dietary cannabinoid. CB2 activation produces anti-inflammatory effects without CB1-mediated psychoactivity. Also a PPAR-gamma agonist. Classified as GRAS (Generally Recognised as Safe) by the FDA as a food flavouring.
Relevance in Cannabis
Typically 5–35% of total cannabis terpenes — one of the most abundant. CB2 agonist activity means BCP may contribute pharmacologically to therapeutic effects of full-spectrum preparations beyond entourage effect hypothesis.
GMP and Manufacturing Relevance
BCP's CB2 agonist status raises potential questions about pharmaceutical classification of high-BCP preparations. In practice, regulatory authorities have not classified BCP as a controlled substance. Include in full-spectrum terpene specifications — its CB2 activity should be noted in product pharmacological profile documentation.
Analytical Measurement
Terpene profiling is performed by headspace GC-MS or GC-FID. Validated methods should achieve quantitative separation of key terpenes. Where terpene content is a product specification parameter, the analytical method must be validated under ICH Q2(R1).
